Purification and Immunological Characterization of beta-hemolysin Produced by Vibrio mimicus


Abstract
The hemolysin produced ty Vibrio mimicus was isolated and purified by ammonium sulfate precifitation, DEAE-sephadex A-50 ion exchange, sephadex G-150 gel filteration chromatography(1st and 2nd) and hydroxyapatite chromatography.
The molecular weight of purified hemolysin was estimated as 58,000 by SDS-PAGE. The pH stability of purified hemolysin was between pH 4.0 and pH 8.0. The hemolytic activity was inactivated by heating 60¡Éfor 5 min. and inhibited by Cu2+, Ni-2+,
Zn2+ and
high concentration of cholesterol.
Immunological cross-reactivity between V. mimicus hemolysin and other Vibrio spp. hemo1ysins was demonstrated by Ouchterlony, neutralization test, ELISA and western blotting assay. These results indicate that the hemolysin from V. mimicus shares
antigenic determinants with hemolysin from V. cholerae non-01.
